Comparison of nematicidal activity of some essential oils and fungal metabolite against Ditylenchus dipsaci under laboratory conditions and identifying their chemical compounds

Abstract

Ditylenchus dipsaci is a plant-parasitic nematode that has a global distribution and is mostly prevalent and causes damage in temperate regions. in this research, the nematicidal activity of herbal essential oils of mountain thyme, oregano, clove, sage, and savory and the metabolite of the fungus Scytalidium sp. were studied in comparison with metam sodium as a positive control under laboratory conditions. To prepare the inoculum of nematode, isolated nematodes from naturally infected garlics collected from Gogan region (East Azarbaijan province), were inoculated on carrot discs and incubated in 18±1°C for 60 days. The bioassays were performed with four repetitions and seven treatments on the reared nematodes and the mortality percent of the nematodes was recorded after 24 and 48 hours. The LC50 values were calculated using probit analysis of the data. The LC50 values after 24 hours for clove, oregano, marzeh and thyme essential oils were 1748, 769.2, 1662 and 4913 ppm, respectively, and 207.6 and 6.7 ppm for fungal metabolite and metam sodium, respectively. Gas chromatography connected to a mass spectrometer (GC-MS) equipped with a DB5 column was used to identify the components of plant essential oils and the metabolite as well. Based on the obtained LC50 values, the tested compounds, except sage-flower, caused the mortality of D. dipsaci in relatively low concentrations and are worth to conduct complementary studies.
