اولین گزارش گونه Pseudoidium neolycopersici عامل سفیدک‌پودری گوجه‌فرنگی بر پایه شناسایی ریخت‌شناختی و مولکولی در ایران

چکیده

سفیدک پودری از بیماری‌های مهم گوجه‌فرنگی در مزارع و گلخانه‌های دنیا محسوب می‌شود و در ایران گونه Leveillula tauricaبه‌عنوان تنها عامل این بیماری گزارش شده است. طی مطالعه قارچ‌های تیره Erysiphaceae در استان اردبیل، برگ‌های گوجه‌فرنگی با علایم سفیدک پودری از گلخانه‌ای در شهر اردبیل جمع‌آوری و جدایه‌ها بر اساس ویژگی‌های ریخت‌شناختی مورد شناسایی اولیه قرار گرفتند. سپس DNA ژنومی قارچ به روش Chelex 5% استخراج گردید و ناحیه ITS از rDNA با آغازگرهای ITS1 و ITS4 تکثیر و توالی‌یابی شد. بر اساس مشخصات ریخت‌شناسی و توالی‌یابی ناحیه ITS-rDNA، این قارچ به عنوان گونهPseudoidium neolycopersici (Syn: Oidium neolycopersici)  شناسایی شد. بدین‌ترتیب، این گونه برای اولین‌بار به‌عنوان عامل سفیدک‌پودری گوجه‌فرنگی (Lycopersicon esculentum) در ایران معرفی می‌شود. گونه P. neolycopersici  یکی از مهم‌ترین عوامل مولد سفیدک‌سطحی گوجه فرنگی در سراسر دنیا است و پیدایش آن در ایران تهدید جدیدی برای کشت گوجه فرنگی در کشور می‌باشد.

چکیده [English]

Powdery mildew is an important disease in the fields and greenhouse-grown tomatoes in the world. Leveillula taurica has been reported as only tomato powdery mildew causal agent in Iran. During study on Erysiphaceae in Ardabil province, tomato leaves with powdery mildew symptoms in one greenhouse in Ardabil city were collected and isolates were identified based on morphological characteristics. Moreover, total DNA was extracted with Chelex 5% method and the ITS region of rDNA were amplified and sequenced using ITS1 and ITS4 primers. According to morphological and molecular characteristics, the fungus identified as Pseudoidium neolycopersici. Thus, this species reported for the first time as powdery mildew on Lycopersicon esculentum in Iran. P. neolycopersici is one of important causal agents of tomato powdery mildew in the world and its occurrence is a new treat on tomato culture in Iran.
